内容記述 Polymeric micelles loading cis-diamminedichloroplatinum(II), CDDP, (cisplatin micelle) has performed higher accumulation in solid tumors and lower toxicities compared to CDDP alone. The combinatorial use of the cisplatin micelle with radiation is expected to enhance the therapeutic effects and to reduce the side effects. The kinetics of the cisplatin micelle, however, has not been fully understood. Particle Induced X-ray Emission (PIXE) has been employed in this study to measure the time transient of platinum in Chinese Hamster Ovary (CHO) cells. The results showed that the platinum content of the cells treated with cisplatin micelle was increased slower than CDDP, suggesting that the cellular uptake could be controlled by the micelle. The released CDDP from micelles were dominantly incorporated into the cells by diffusion. The uptake characteristics were analyzed further by using the micelles with different collapse rates. These results and techniques used in this study will be useful for designing an optimum treatment plan for the combination of platinum-containing polymeric micelles and radiation toward clinical application.
