内容記述 | Hemoglobin is a major protein in the erythrocytes of vertebrates and transports oxygen to various organs and tissues in the body. Hemoglobin is a tetramer protein consisting of two alpha- and two beta-globin polypeptides. In vertebrates, it is well known that alpha- and beta-globins are switched from embryonic to adult type during development. We characterized the embryonic globins in medaka, and determined the full-length structure of medaka globin gene locus. Medaka had two globin gene clusters: One consisted of 9 embryonic globin genes and mapped to linkage group (LG) 8, the other comprised 3 adult globin genes and mapped to LG 19. These data indicate that the genomic organization of globin genes of medaka differs from those reported in higher vertebrates such as mammals, birds and amphibians. Analysis using RT-PCR showed that the developmental expression patterns of 11 genes were classified into four types. We selected a typical gene from each of the four types, fused its 5'-upstream region including probable minimum promoters with a reporter gene, and introduced the fusion gene into embryos. Thus, we established 4 types of transgenic lines stably expressing the reporter gene in erythroid cells. Each transgenic medaka expressed the reporter gene in a developmental stage-specific manner. Compared with locus control regions in mammals, a relatively shorter 5'-upstream cisregulatory region had the ability to promote or enhance the developmental stage-specific expression of medaka globin genes. |
