| 内容記述 | We developed an AFLP marker detection technique using capillary gel electrophoresis to confirm the presence or absence of mutations and to detect chimeric mutation in ion-beam irradiated Japanese pear cultivars. First, we evaluated the reliability of this method by identifying six varieties of Japanese pear. A total of 180 peaks were obtained, of which 103 were variety-specific. A phylogenetic tree based on the peaks obtained was consistent with the variety's genealogy, confirming the method's reliability. A total of 190 peaks were obtained between the ion-beam irradiated and non-irradiated individuals, of which 35 were mutant-specific peaks. Different peaks were detected from different leaves of the same individual in ion-beam irradiated plants, allowing the detection of chimeric mutation. Furthermore, the chimeric mutation rate was found to be reduced in the shoots of the two-year-old branch via the lateral buds. | |||||||||||
