Aims In this study, we investigate the cell killing effects of carbon-ion beam IR combined with multikinase inhibitor sorafenib on hepatocellular carcinoma (HCC) cells.Materials & Method HCC cells and cancer stem cells (CSCs) sorted from HepG2 and Huh7 were treated with carbon-ion beam irradiation (IR) alone or in combination with sorafenib, a multikinase inhibitor, then cell viability, colony and spheroid formation ability, cell death- and angiogenesis-related gene expression changes were analyzed.Results The combination of carbon-ion beam IR with sorafenib (10uM) significantly suppressed HCC cell viability and remarkably decreased colony and spheroid formation ability in CSCs (CD133+/CD90+, CD44+/ESA+) isolated from HepG2 cells compared to carbon-ion beam IR or X-ray IR alone. Annexin V and caspase3/7 activity assay revealed that the combination of carbon-ion beam IR and sorafenib significantly induced apoptosis compared to that of its alone treatment. Custom PCR array system analysis showed that carbon-ion beam IR combined with sorafenib significantly enhanced expressions of Bax and Cytochrome c and decreased HIF1a and VEGF.Conclusions The combination treatment of carbon-ion beam IR and sorafenib significantly destroyed HCC cells including CSCs accompanied with apoptosis induction. |
