Radiation exposure in the period leads to disturbances in development of the brain and can result in microcephalia and other neurological dysfunctions such as mental retardation. We have demonstrated that X-irradiation of the NSCs, prepared from mouse embryonic stem (ES) cells by the NeuralStem Sphere method, inhibits proliferation dose-dependently. In this study, we investigated effects of X-irradiation of NSCs on proliferation and differentiation into the neural cells. When proliferating NSCs were irradiated with 1 Gy, the cells stopped proliferation but started to proliferate exponentially with a lag time of about 1 day. The cells expressed nestin and maintained potential of proliferation and differentiation like non-irradiated cells. When the NSCs were irradiated with 5 Gy, the cells stopped proliferation and became larger in size. The cells expressed nestin, however, lost potential of proliferation and differentiation.
