Here, we take advantage of bacterial\ndihydrofolate reductase, ecDHFR, and its unique antagonist, TMP,\nto facilitate in vivo imaging in the brain. Peripheral administration\nof radiofluorinated and fluorescent TMP analogs enabled PET and\nintravital microscopy, respectively, of neuronal ecDHFR expression\nin mice. This technique can be used to the visualize neuronal\ncircuit activity elicited by chemogenetic manipulation in the\nmouse hippocampus. Notably, ecDHFR-PET allows mapping of\nneuronal projections in non-human primate brains, demonstrating\nthe applicability of ecDHFR-based tracking technologies for\nnetwork monitoring. Finally, we demonstrate the utility of TMP\nanalogs for PET studies of turnover and self-assembly of proteins\ntagged with ecDHFR mutants. These results establish opportunities\nfor a broad spectrum of previously unattainable PET analyses of\nmammalian brain circuits at the molecular
