Carbon ions (C-ions) act independently of the oxygen concentration and trigger complex- and clustered-DSBs difficult to repair. Understanding the interrelation between hypoxia, radiation-type, and DNA-repair is therefore essential for overcoming radioresistance. The DSBs signaling and the contribution of the canonical non-homologous end-joining (NHEJ-c) and homologous-recombination (HR) repair pathways were assessed by immunostaining in two cancer-stem-cell (CSCs) and non-CSCs HNSCC cell lines. Detection and signaling of DSBs were lower in response to C-ions than photons. Hypoxia increased the decay-rate of the detected DSBs (γH2AX) in CSCs after photons and the initiation of DSB repair signaling (P-ATM) in CSCs and non-CSCs after both radiations, but not the choice of DSB repair pathway (53BP1). Additionally, hypoxia increased the NHEJ-c (DNA-PK) and the HR pathway (RAD51) activation only after photons. Furthermore, the involvement of the HR seemed to be higher in CSCs after photons and in non-CSCs after C-ions. Taken together, our results show that C-ions may overcome the radioresistance of HNSCC associated with DNA repair, particularly in CSCs, and independently of a hypoxic microenvironment.","subitem_description_type":"Abstract"}]},"item_8_relation_14":{"attribute_name":"DOI","attribute_value_mlt":[{"subitem_relation_type_id":{"subitem_relation_type_id_text":"10.1038/s41598-020-78354-7","subitem_relation_type_select":"DOI"}}]},"item_8_relation_17":{"attribute_name":"関連サイト","attribute_value_mlt":[{"subitem_relation_type_id":{"subitem_relation_type_id_text":"https://www.nature.com/articles/s41598-020-78354-7","subitem_relation_type_select":"URI"}}]},"item_8_source_id_9":{"attribute_name":"ISSN","attribute_value_mlt":[{"subitem_source_identifier":"2045-2322","subitem_source_identifier_type":"ISSN"}]},"item_access_right":{"attribute_name":"アクセス権","attribute_value_mlt":[{"subitem_access_right":"metadata only access","subitem_access_right_uri":"http://purl.org/coar/access_right/c_14cb"}]},"item_creator":{"attribute_name":"著者","attribute_type":"creator","attribute_value_mlt":[{"creatorNames":[{"creatorName":"Anne‑SophieWozny"}],"nameIdentifiers":[{"nameIdentifier":"1006985","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Alphonse, Gersende"}],"nameIdentifiers":[{"nameIdentifier":"1006986","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Cassard, Audrey"}],"nameIdentifiers":[{"nameIdentifier":"1006987","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Céline, Malésys"}],"nameIdentifiers":[{"nameIdentifier":"1006988","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Louati, Safa"}],"nameIdentifiers":[{"nameIdentifier":"1006989","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Beuve, Michael"}],"nameIdentifiers":[{"nameIdentifier":"1006990","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Lalle, Philippe"}],"nameIdentifiers":[{"nameIdentifier":"1006991","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Ardail, Dominique"}],"nameIdentifiers":[{"nameIdentifier":"1006992","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Nakajima, Tetsuo"}],"nameIdentifiers":[{"nameIdentifier":"1006993","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Claire, Rodriguez‑Lafrasse"}],"nameIdentifiers":[{"nameIdentifier":"1006994","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Tetsuo, Nakajima","creatorNameLang":"en"}],"nameIdentifiers":[{"nameIdentifier":"1006995","nameIdentifierScheme":"WEKO"}]}]},"item_language":{"attribute_name":"言語","attribute_value_mlt":[{"subitem_language":"eng"}]},"item_resource_type":{"attribute_name":"資源タイプ","attribute_value_mlt":[{"resourcetype":"journal article","resourceuri":"http://purl.org/coar/resource_type/c_6501"}]},"item_title":"Impact of hypoxia on the double-strand break repair after photon and carbon ion irradiation of radioresistant HNSCC cells","item_titles":{"attribute_name":"タイトル","attribute_value_mlt":[{"subitem_title":"Impact of hypoxia on the double-strand break repair after photon and carbon ion irradiation of radioresistant HNSCC cells"}]},"item_type_id":"8","owner":"1","path":["1"],"pubdate":{"attribute_name":"公開日","attribute_value":"2021-03-09"},"publish_date":"2021-03-09","publish_status":"0","recid":"82094","relation_version_is_last":true,"title":["Impact of hypoxia on the double-strand break repair after photon and carbon ion irradiation of radioresistant HNSCC cells"],"weko_creator_id":"1","weko_shared_id":-1},"updated":"2023-05-15T19:18:06.327995+00:00"}
