In order to separately examine repairability\nof DNA damage from damage responses of organelles,\nEGFP-expressing plasmid DNA (in TE buffer) was\nexposed to X-rays in vitro, and then transfected into \"\nNON-irradiated \" human breast cancer cells (MCF7). The\ndynamics of EGFP fluorescence of cells was observed\nby a live cell imaging technique to analyze DNA repair\nefficiency in the cells. The EGFP expression kinetics as\nDNA repair efficiency was analyzed for the cells transfected\nwith the plasmids exposed to several X-ray doses.\nThe similar experiments were also performed using\nplasmids treated with nicking or restriction enzyme to\nobtain positive control data of SSB or DSB induction,\nrespectively. Based on those positive control values,\ntheoretically expected efficiencies were also calculated\nassuming that strand breakage of plasmid DNA by irradiation\nfollowed Poisson distribution. The numbers of\ncells expressing EGFP in the experimental results were\nconsiderably fewer than the theoretical values, indicating\nthat the irradiated plasmids were less repaired in the\ncells.
