To understand how the torsional stress changes the stability of nucleosomes, we carried out all-atom molecular dynamics simulations to examine the unwrapping of the outer superhelical turn of the nucleosomal DNA from the histone octamer under the torsional stress imposed on the edges of the DNA. We found that the torsional stress can control the transitional path of symmetric and asymmetric unwrapping of the DNA by changing the interaction between the DNA and the histone core through conformational change in the DNA.
