It delivers a focused vertical microbeam of 3.4 MeV protons to a targeted cell nucleus or cytoplasm. Such advantages of the SPCIE have motivated further investigations on the cellular response to different DNA damage complexities induced by ionizing radiation. Previously, we reported an accurate and systematic investigation of the SPICE microbeam profile using a FNTD [2]. In the present study, as a second step, we discuss the localized dose distribution in a targeted nucleus of human lung normal WI-38 cells and its correlation with intensity and area of DNA double strand break (DSB) region, visualized by immunostaining against g-H2AX. Finally, superlinear dependence of the g-H2AX spot area to the beam area was interestingly observed for large number of protons per position, suggesting that the penumbra dose fraction in the localized dose distribution significantly contributed to the DSB induction.
