SQAP が in vivo のみで誘発する放射線増感効果の作用メカニズムを、腫瘍の血管新生や酸素動態などの生理学的特性に着目して調べた。\n【方法】SQAP (2 mg/kg) を SCCVII あるいは A549 担癌マウスへ尾静注し、必要に応じて X 線 (2 Gy) を照射後、電子スピン共鳴酸素イメージング、光音響画像解析、Gd-DTPA を用いた DCE-MRI (T1) および微小超常磁性酸化鉄粒子 (USPIO) を用いた MRI (T2) により、腫瘍内酸素分圧 (pO2)、血中酸素飽和度 (sO2)、腫瘍内還流、微小血管密度の変化を経時経日的に追跡した。\n【結果】尾静注された SQAP は、①赤血球からの酸素分子の放出促進作用 (sO2 低下: ~15%) および②腫瘍組織内の還流上昇作用 (~110%) を示すことが判明し、投与 20-30 分後には腫瘍内 pO2 が最大で 7.8 mmHg 上昇した。このタイミングで X 線を照射すると、その酸素効果が増大し、治療効果が増強される可能性が示唆された。また、併用治療を 3 日間毎日継続した後の腫瘍では、その生育速度が遅延するとともに、低酸素化の抑制、微小血管の消失、血管周皮細胞の一過的発現など、腫瘍血管の正常化 (Vascular normalization) を示唆する所見が認められた。\n【結論】各種 in vivo 分子イメージングにより腫瘍内微小環境の生理学的特性を多角的に評価し、in vitro 実験では解析困難な生理学的変化に起因する SQAP の放射線増感作用のメカニズムを解明した。含硫糖脂質としては初の知見となる本生物活性を基盤に、GLP 準拠の製剤化 (Lavurchin®, M.T.3 社)
