Three nitroxyl contrast agents, 3-carboxy-2,2,5,5-tetramethylpyrrolidine-N-oxyl (CxP), 3-carbamoyl-2,2,5,5-tetramethylpyrrolidine-N-oxyl (CmP), and 4-hydroxy-tetramethylpiperidine-N-oxyl (TPL), having different lipophilicities were compared using MR redox imaging. The MR signal decay rate of CxP was slower in tumor tissue than normal tissue, while the decay rates of CmP and TPL were faster in tumor tissue than normal tissue. Exposure to 100% oxygen made decay rate of TPL in tumor tissue faster, but not significantly. No marked effect was obtained on decay rate of TPL in normal muscle. In addition, 100% oxygen challenging make maximum T1 enhancement induced by TPL in tumor tissue lower but not significant. Oxygen challenges gave no marked effect on the nitroxyl-induced maximum T1 enhancement in the normal tissue. Breathing 100% oxygen could make the concentration difference of TPL between tumor and normal tissues larger. The EPR linewidth of TPL in normal tissue was increased by Carbogen breathing, while 100% oxygen breathing reduced the EPR linewidth of TPL.
