SLE\npatients exhibit the increased number of CD180-negative B cells in blood,\nwhich appears to be associated with the activity of SLE. The production of\nautoantibodies was found on CD180-negative cells from SLE patients. In\nSLE-model NZBWF1 mouse, we previously reported the increased number\nof CD180-negative B cells with development of SLE morbidity and the\nproduction of autoantibodies by CD180-negative cells. We also showed\nCD180-negative cells were more radiosensitive than CD180-positive cells.\nThese suggest that radiation may lead to selective elimination of CD180-\nnegative cells, resulting in a decrease in the production of autoantibodies\nin SLE. We therefore attempted to examine the effects of X-ray irradiation\non CD180-negative cells in NZBWF1 female mice. After the onset of\nsymptoms, mice were paired according to their severity. In each pair, one　\nmouse was irradiated by 4 Gy sealing her head, and the other was shamirradiated.\nCompared to the sham-irradiated mice, the mean number of\nCD180-negative cells in spleen from irradiated mice was significantly\ndecreased, and the mean life span was markedly extended. The latency to\nthe development of grade three-proteinuria in irradiated mice is also\nremarkably greater than that in sham-irradiated mice. Within each pair,\nthe life span of the irradiated mice was significantly longer than that of the\npaired sham-irradiated mice. These suggest that radiation could effectively\nelim inate CD180-negative B cells, leading to reduction of autoantibody\nproduction, and consequently relieve the SLE symptoms in mouse.
