Exposure of the fetal brain to ionizing radiation cause congenital brain abnormalities, which is thought to result from defects in neurogenesis during the developing stage. Here, we investigated the effects of X-irradiation on proliferating homogenous NSCs, which were prepared from mouse embryonic stem cells by the Neural Stem Sphere method. Cells cultured in the proliferation medium, containing fibroblast growth factor-2 (FGF-2) were irradiated with X-rays and analyzed with growth curve measurements, flow cytometry, real-time RT-PCR, and immuno-fluorescence. The cells irradiated at a dose of > 5 Gy did not show an increase in number, but a gradual decrease was observed. The gradual decrease was explained by the loss of proliferation capability and also apoptosis. In contrast, the cells irradiated at a dose of 1 Gy stopped proliferation temporarily but recovered the proliferation capability. Moreover, sub-cultured at 96 hrs after irradiation, the cells proliferated logarithmically in the proliferation medium, and successfully differentiated into neurons and astrocytes in the differentiation medium. These results clearly demonstrate that exposure of the fetal brain to radiation at relatively high doses leads to congenital morphological and functional brain abnormalities.
