Previously, we have shown that gap junction intercellular communication (GJIC) mediates the propagation of stressful effects in irradiated cells exposed to high linear energy transfer (LET) radiations (alpha particles or iron ions) by which every cells in the population is traversed by IR track.1-2 Here, we have pursued these studies and investigate the role of GJIC in modulating the repair of potentially lethal damage (PLDR) and micronuclei formation, a reflection of DNA damage, in human cells exposed to low- or high-LET radiations at Heavy Ion Medical Accelerator in Chiba (HIMAC) in Japan. \nConfluent normal human skin fibroblasts, which were obtained from the Riken BioResource in Japan (NB1RGB: Cell No. RCB0222) were exposed in the presence or absence of gap junction inhibitor alpha-glycyrrhetimic acid (AGA) to 3.7 Gy from 200 kV X rays (LET~1.7 keV/um), 1.1 Gy from carbon ions (LET~76 keV/um), 1.2 Gy from silicon ions (LET~113 keV/um) or 1.3 Gy from iron ions (LET~400 keV/um that result in isosurvival levels. They were incubated for various times (5-10 min, 3 h or 24 h) at 37C. As expected, high-LET radiations were more effective than low-LET radiations at inducing of cell killing and DNA damage within 5-10 min after irradiation. However, when cells were held in confluent state for several hours, PLDR coupled with a reduction in DNA damage, occurred only in cells exposed to low LET X rays, but not in those exposed to high-LET radiations (carbon ions, silicon ions or iron ions). Interestingly, inhibition of GJIC by AGA eliminated the enhancement of toxic effects which correlated with an increase of cell survival and a reduction in the level of micronucleus formation in cell cultures exposed to high-LET radiations but not in low-LET X rays. \nTogether, these results show that radiation quality (LET) and GJIC play an important role of the propagation of stressful effects in among irradiated confluent cells exposed to high-LET radiations and that GJIC has only minimal effect on PLDR and induction of DNA damage following low-LET radiations.
