The aim of this study is to investigate the underlying mechanisms of radioresistance and the effectiveness of carbon ion (C-ion) irradiation on the radioresistant cancer cells. To establish radioresistant cancer cell lines, we repeatedly irradiated a mouse squamous cell carcinoma cell line, NRS1, with X-ray up to 60 Gy. The repetitive X-ray irradiation altered the shape of the NRS1 cells, and their size was approximately 2-folds larger than that of non-irradiated cells. Subsequently, ten clones were selected as radioresistant cancer cell lines. These clones could be categorized into the following three groups; (1) the shape is approximately the same as the majority of irradiated NRS1, (2) have lost cell-to-cell contact and are spindle-like, and (3) undifferentiated-like form.
