However, the regulation of gene expression by DNA methylation after genotoxic stress, including irradiation, remains poorly understood. We hypothesized that irradiation may elicit alterations of DNA methylation status and subsequently cause gene expression.\n In this study, we have attempted to integrate the results of comprehensive gene expression analysis and DNA methylation analysis in cellular response to irradiation in MIAPaCa-2 and PANC-1 cell lines, both derived from human pancreatic cancer. Gene expression in these cell lines at 48 h after irradiation with X-ray at 4 Gy were analyzed by microarray in which 27,958 targets from Entrez Gene RNAs were designed as probes. DNA methylation status in the cell lines in these conditions were analyzed by next generation sequencing. Genome regions showing different methylation status between before and after irradiation were detected using MACS algorithm.\n In total, 98 genes changed expression after irradiation in these conditions. Genome regions ranging from 30 Mb to 50 Mb were methylated or de-methylated in MIAPaCa-2 or PANC-1 cell lines after X-ray irradiation. As expected, alterations of DNA methylation status by irradiation in approximately 150 regions were found within the 2 kb of the 45 genes that changed their expression after irradiation. Studies are in progress to examine the relationships between gene expression and alteration of DNA methylation status in cells after irradiation.
