This necrosis-regeneration cycle would in theory be linked to neoplastic transformation of normal mucosal epithelia. In this study, six-week-old C57BL/6J male mice were induced ulcerative colitis by administering 1 or 2% dextran sodium sulfate (DSS) for 7 days. Up until 3 weeks after DSS treatment, we examined sequentially the mucosal regeneration process histopathologically. Mucin-depleted foci (MDF), which are considered as precancerous lesions of colon carcinoma, were recognized just after completion (day 0) of DSS administration in the adjacent ulcerated lesions. Aberrant crypt foci-like lesions (ACF-L) were observed at day 2 after DSS administration in the same area (as MDF). The lesions having characteristics of both MDF and ACF-L showed the atypical changes with nuclear stratification, loss of nuclear polarity and structural abnormality of the crypts.
