Radiocarbon (14C) is one of the dominant radionuclides contributing to the dose from TRU waste. Recently, it was found that 14C atoms in organic matter such as carboxylic acids are released from metallic TRU waste. In our previous study using [1,2-14C] sodium acetate, we found that bacteria are responsible for the behavior of 14C atoms in organic matter in Japanese paddy soils. In addition, bacteria that existed in deionized water used for flooding of agricultural soils in our batch culture experiments were found to utilized sodium acetate. The objectives of the present study are to isolate the acetate-utilizing bacterium and to identify the species name. R2A agar plates containing [1,2-14C] sodium acetate were used to isolate the acetate-utilizing bacterium. Autoradiography images showed that all colonies on the R2A plates were positive for 14C utilization. Then we isolated one of the colonies. A partial sequence of 16S rRNA gene was used for the identification of the bacterium. Results of the sequence indicated that the bacterium had high homology (99.4%) to Burkholderia gladioli. B. gladioli is found in soil, water, and the rhizosphere. Bacteria that affect the behavior of 14C atoms in organic matter will be omnipresent in agricultural soils.
