To identify biochemical alternations that would be the markers in human mesothelioma, we measured the intracellular steady-state level of biologically important trace metals such as manganese (Mn), copper (Cu), zinc (Zn) in a human mesothelial cell line (MeT-5A) and in five human mesothelioma cell lines by inductively coupled plasma-mass spectrometry (ICP-MS). Furthermore, we determined the intracellular levels of manganese-SOD (Mn-SOD) and copper/zinc-SOD (Cu/Zn-SOD) to investigate the relationship between their enzymatic activities and the metal contents. Some mesothelioma cells had significantly higher amounts of Mn or Cu than MeT-5A. Interestingly, all mesothelioma cells highly expressed Mn-SOD compared with MeT-5A. The cells with increased Mn-SOD activity contained higher amounts of Mn, suggesting that intracellular Mn content is positively correlated with Mn-SOD.
