Overexpression of c-kit with its functional mutation is present in most GISTs. Positron emission tomography using 18F-fluoro-deoxyglucose (FDG-PET) has been useful for the detection of GIST and for the assessment of response to Imatinib treatment. However, FDG-PET, in some cases fails to detect relapse of GIST at early stage. If we could develop an imaging method specific to c-kit expression, it can be used for the evaluation of the early relapse of GISTs. In this study, we radiolabeled anti-c-kit monoclonal antibodies (Mabs) with 125I and 111In, and assessed their in vitro and in vivo characteristics. We radiolabeled Mabs with 125I and 111In, and performed cell binding, competitive inhibition and internalization assays in vitro, and biodistribution and SPECT imaging in vivo using tumor-bearing mice. These results showed that radiolabeled Mabs specifically bound to c-kit expressing cells and were internalized, and 111In labeled Mab accumulated in tumors and visualized tumor by SPECT.
