The careful design of the architecture of water-soluble QDs enables the creation of nanobioprobes that are compatible with several imaging techniques, so-called multimodal imaging. This opens the door to the existing prospect of simultaneous application of QD probes in a variety of techniques including optical imaging, magnetic resonance imaging (MRI), positron emission tomography, computed tomography and X-ray imaging.\nIn the present study, we describe two multimodal QD probes with combined fluorescent and paramagnetic properties (Figure 1) and their simultaneous application in optical imaging and MRI. Using fluorescent confocal microscopy, we observed that the probes were characterized with easy and comparatively fast intracellular delivery in viable cells without transfection techniques that are inherently toxic or keep a risk of artifacts. Both probes penetrated through cytoplasmic membrane via endocytosis. The probes were localized into the cytoplasm, uniformly distributed. Using MRI, we quantified the amount of multimodal QDs delivered into the cells or adsorbed on the cell surface. Two independent calibration curves were used, based on the relationship between the concentration of the probe and T1 (longitudinal relaxation time) or T2 (transverse relaxation time) values, respectively. It was found that approximately 58% of the probes were delivered into the cells and/or bound on the cell surface. The photosensitization and cytotoxicity were clarified on several cultured cell lines before and after laser or ultraviolet irradiation. No cytotoxicity or photosensitization was detected up to 500 nM concentration.\nFigure 1.
