Thus, it is important to develop more effective treatments. Noninvasive imaging is essential for assessment of the efficacy of new treatments. To establish noninvasive imaging of mesothelioma, we compared the tumor uptake and biodistribution of three types of PET tracer, [F-18]-FDG, [F-18]-FLT and [C-11]-thiothymidine, in mesothelioma model mice. We inoculated two histological subtypes of mesothelioma cell lines, epithelioid and sarcomatoid, into subcutaneous tissue and the thoracic cavity of nude mice, and established subcutaneous and orthotropic tumor models. For biodistribution or PET imaging, mice were administrated with FDG, FLT or thiothymidine, and 50 min later, we measured the radioactivity of tissues and tumors, or conducted a 10-min emission scan. Two thymidine analogue, FLT and thiothymidine, highly accumulated in epithelioid mesothelioma, while a glucose analogue, FDG, highly accumulated in sarcomatoid mesothelioma. Then, to measure in vitro cellular uptake of FDG and FLT of each cell line, we incubated cells with 150nmol tracers at 37DegreeC, and then measured the radioactivity of cell at several time points. Moreover, to measure the cytosolic thymidine kinase 1 (TK1) activity of each cell line, we isolated cytosolic fraction by ultracentrifuge, incubated cytosolic proteins with 1pmol [H-3]-thymidine at 37DegreeC, and then measured the radioactivity of phosphorylated thymidine at several time points. The cellular uptake of both FDG and FLT in sarcomatoid mesothelioma was higher than that in epithelioid mesothelioma, and the TK1 activity of sarcomatoid was higher than epithelioid. These results indicated that in vitro characters are not always consistent with in vivo behavior.
