Normal human fibroblast cell line AG1521 was treated with the inhibitor cocktail at various time points AFTER irradiation, and the number of foci was measured up to three hours after addition of the mixture. When the inhibitors were added at 1hour after irradiation, no change in the number of foci was observed, but the addition of the mixture at 6, 12, 24 hours after irradiation significantly decreased the number of foci. The background level of foci without irradiation was also reduced with the kinase inhibitors within three hours and this reduction was not observed when the mixture were treated with 4C. Interestingly, the level of foci number returned to the background level when cells were cultured in the inhibitor free medium. Our findings indicate that the number of foci in the background level and in the residual level after irradiation was derived from the equilibrium state between kinase and phosphatase activities.
