Especially in our body 15-deoxy-delta 12,14-prostaglandin J2 (15d-PGJ2) is produced and acting as a cytoprotector. We examined whether 15d-PGJ2 activates heme oxygenase-1 (HO-1) gene in mouse monocyte-macrophage cell line, RAW 264.7. Cells were treated with 15d-PGJ2 for 4 h in DMEM. mRNAs of HO-1 and GAPDH were estimated by the RT-PCR method. As a result we detected a 105 times increase of HO-1 mRNA with 500 nM 15d-PGJ2. We have investigated whether PPARgamma is involved in this activation. At first we examined the effect of troglitazone which is an agonist of PPARgamma. We could not detect any increase of HO-1 mRNA by troglitazone. Therefore it was confirmed that the activation of PPARgamma does not activate the transcription of HO-1 gene. We next examined the effect of GW9662, which is an antagonist of PPARgamma, upon transcription of HO-1 gene. GW9662 did not affect the mRNA of HO-1 gene.
