Incubation with added L-Tyr inhibited the proliferation of both Slt/Slt and slt/slt melanocytes in a concentration-dependent manner and that inhibition was gradually augmented as the donor mice aged. In contrast, the differentiation of melanocytes in slt/slt mice was inhibited compared to that in Slt/Slt mice. Tyrosinase-related protein (TRP)-1, TRP-2 and c-kit were expressed in the slt/slt melanocytes, though the expression of TRP-2 in the slt/slt melanocytes was reduced to 30-50% than that in Slt/Slt melanocytes. L-Tyr induced the differentiation of slt/slt melanocytes as the age of the donor mice advanced. The content of the eumelanin marker, pyrrole-2,3,5-tricarboxylic acid in slt/slt melanocytes was reduced compared to that of Slt/Slt melanocytes. However, the content of the pheomelanin marker, 4-amino-3-hydroxyphenylalanine in slt/slt melanocytes was similar to that of Slt/Slt melanocytes. L-Tyr increased the content of eumelanin and pheomelanin 2- to 3-fold in cultured cells derived from 0.5-, 3.5- and 7.5-day-old mice both in Slt/Slt and slt/slt. However, the content of eumelanin and pheomelanin in culture media derived from slt/slt mice was greatly increased after treatment with L-Tyr, especially from 7.5-day-old mice.
