In the absence of adequate geetic analysis at the molecular level, we have investigated the changes in genetic structure caused by the irradiation of heavy charged particles in cell stains from various types of malignant tumor tissue in the head and neck. Four types of cell lines, namely aquamous cell carcinoma(Ca9-22), malignant melanoma(G361), salivary gland carcinoma (HSG) and keatinocyte (HaCaT), were analyzed by irradiation with heavy charged particles (carbon ion radiation)(1, 4, 7GyE) and photons (1, 4, 8Gy).After irradiation, DNA was extracted (after 1, 24, and 48 hours) for analysis of the 6 microsatellite regions chromososome 17 using the PCR-LOH assay. After photon irradiation, LOH was extensively found at low radiation doese in squamous cell carcinoma cells after 48 hours and in adenocarcinoma cells after 24 and 48 hours. At high doses, however, LOH incidence was lower in all cell types. After heavy charged oartcle irradiation, an LOH pattern was detected only in one malignant melanoma site, with all other lesions showing loss of homozygosity. This indicates that heavy charged parrticles induse lethal changese in the DNA regardless of time and dose.
