Our recent investigation indicated that 17AAG, an effective Hsp90 inhibitor derived from geldanamycin, clearly gave radiosensitizing effect in SQ5 human lung tumor cells irradiated with carbon ions. In this study, another Hsp90 inhibitor PU-H71 was examined for the combined effect with carbon ions. Our data indicated that pretreatment of 1 uM PU-H71 clearly showed an enhanced radio-sensitization in SQ5 cells irradiated with carbon ions (290 MeV/n, ~ 50 keV/um at spread-out Bragg peak). Our preliminary in vivo mouse xenogratft study also revealed an advantage in tumor growth control in the combined treatment of PU-H71 (40mg/kg) and carbon ion irradiation when compared to carbon irradiation alone. The cause of this radio-sensitizing effect by PU-H71 is under investigation; however, an effect on Rad 51, a DNA Double strand break (DSB) repair protein, was clearly observed in tumor cells treated with PU-H71 and carbon ion irradiation.
