Here, we explored the feasibility of TSPO as an imaging biomarker in the detection of hepatocellular carcinoma (HCC) that could be monitored using PET with a TSPO radioligand [18F]FEDAC. Cell binding assay and PET/CT with [18F]FEDAC quantified TSPO in HCC cell lines differing in their invasive and chemotactic potential and orthotopic HCC. Uptake of [18F]FEDAC was increased in the highly aggressive cell lines HLE (12.07 +/- 0.96 %ICD/mg protein), relative to nonaggressive cell line HepG2 (3.87 +/- 0.36 %ICD/mg protein). Uptake of radioactivity was 3.71 fold in the livers of orthotopic HCC compared with the control mice ( p<0.05 ). Conclusions: TSPO expression increases in HCC and closely correlates with HCC aggressive phenotype expression.
