Mouse cultured and frozen ES cells (-80 degree in C) were irradiated by X-ray, Fe and C ion heavy particles. The cells were plated and numbers of colony were counted. The mouse ES cells were most sensitive to Fe ion particles at 0-1Gy of exposure and carbon gave moderate effect to ES cells. Interestingly, radiation at low dose of 0-1 Gy, by Fe and C particles damaged ES cells seriously, while X-ray did not We also examined phosphorylation of histone H2AX after irradiation of X-ray, Fe particles.
