CD34+ cells derived from placental and umbilical cord blood were induced by carbon-ion beams (LET = 50 keV) or X-rays at 2 Gy, differentiated into megakaryocytes and platelets with thrombopoietin (TPO) and interleukin-3 (IL-3) for 7 days and then assayed using real time RT-PCR. FLI1 and RUNX1, positive regulators of megakaryocytopoiesis, were up-regulated by the exposure to carbon-ion beams. The expressions of MPL, ITGA2B and GPIBA, the megakaryocyte-specific genes, were reduced by X-irradiation. In other factors, X-rays increased the expression of KIT, the receptor for SCF while carbon-ion beams enhanced the expression of C/EBPA, a myeloid transcription factor. These results showed that the regulation of several gene expressions were dependent on each radiation, respectively.
