We previously demonstrated the existence of AR in mice during\nlate organogenesis. For better understanding of molecular mechanisms\nunderlying AR in this model, we performed a global analysis of\ntranscriptome regulations in cells collected from whole mouse fetuses,\nafter in utero exposure to priming irradiation. Several combinations\nof radiation dose and dose-rate were applied to induce or not an AR in\nour system. Expression levels of 14000 different genes 24 hours after\nirradiation were measured. While many genes were not modulated at all\nafter radiation exposure and 166 genes were deregulated for all\nconditions, we identified a panel of 861 genes (so called \"AR genes\")\nshowing significantly different expression ratios under AR-inducing\nand non AR-inducing conditions. A sub-list of 119 AR genes (so-called\nARS genes) with higher discriminative characteristics was also\ncreated. We assessed the proportion of total and AR genes involved in\nvarious molecular pathways and the number of these genes for each of\nthe GO terms. Gene enrichment analysis revealed that growth factor,\ntranslation or metallopeptidase activities (among others) were\npotential targets for our study. Since AR genes were involved in a\nvariety of functions and cellular processes, we applied a functional\nclassification algorithm which clustered genes in a limited number of\nfunctionally related groups. Our results emphasized the role of signal\ntransduction mechanisms in the induction of AR (one-third of AR genes\nwere directly involved in cell signaling). Previous investigations\nsuggested the crucial role of p53 in cells's response to radiation and\npotentially AR; we observed that a significant proportion of AR genes\nwere p53-related or possessed p53-binding sites. Furthermore, many AR\ngenes were important for fetal development, like growth factors or\ngenes involved in signaling cascades (CK2 ...). Taken together, our\nresults present the first evidence of molecular mechanisms modulation\nby AR in utero, which may have potentially important consequences for\nfurther developmental processes, and could even influence\ntumorigenesis.
