In the first year of this study, aiming to irradiate the lung or the urinary bladder of mice with carbon-ion, new type of collimators was constructed. Materials and methods --- C3H/HeNrs and C57BL/6J, 8-week-old female mice were used through experiments. The position of the target, either lung or bladder, was confirmed by x-ray photographs. Also, the absorbed dose at the target was confirmed using photoluminescence dosimeter (PLD) and ionization chamber. Exposures were conducted using horizontal carbon-12 beams with 290 MeV/u at a dose rate of approximately 3 Gy/min within the 6-cm spread-out bragg peak (SOBP). Gamma-rays were used as reference beam with new collimators. To evaluate the effects of C-ion irradiation on lungs, pathological changes and immunohistochemical stainings for CD44 and Bak at 6 hr, day16, 28 and 56 after thoracic irradiation were analyzed. To evaluate the adverse effects on the bladder,cystometry and pathological evaluation were performed.The total RNA was extracted from irradiated tissues, which were removed on day1,4,8,15,29,60,90,150,240 and 360 after irradiation for microarray analysis to elucidate the molecular response to irradiation. Results --- The number of CD44 and Bak positive cells in the irradiated lungs were changed after irradiation.
