The new term nanomedicine has been used recently. Employing constructs such as quantum dots, dendrimers, liposomes, fullerenes, nanoshells, and emulsions, these advances lead toward the concept of personalized medicine and the potential for very early, even pre-symptomatic diagnosis coupled with highly effective targeted therapy.1 The nanoparticles allow concentration of a large amount of contrast substances in a very small area/volume (~20-100 nm) and increasing of sensitivity of molecular imaging techniques. Moreover, if nanoparticles are conjugated with target-specific ligands, even single cells could be visualized in the blood stream or tissues at comparatively low concentration of the probe.\nIn the present study, we describe a development of several original molecular probes: (i) based on multimodal nanoparticles (silica spheres embedded with photoluminescent quantum dots and paramagnetic substances), conjugated with target-selective ligands, or (ii) based on chemically modified spin-labels (stable nitroxyl radicals). All probes possess multimodal spectral characteristics. Some of them could be applied simultaneously in optical imaging and MRI, and others could be applied simultaneously in EPR imaging and MRI.\nAll probes were tested for toxicity on viable cells and/or animals. The nanoprobes were conjugated with biocompatible ligands and applied in the imaging of different molecular targets on cultured cells, using fluorescent confocal microscopy and MRI. Spin-labeled probes were applied for detection of pharmacodynamic of conventional drugs in blood in vivo using EPR spectroscopy, as well as for non-invasive evaluation of permeability of conventional drugs for blood-brain barrier using MRI. \n\\n1Bakalova.
