Colony and spheroid formation assay as well as in vivo tumorigenic assay showed that CD44+/ESA+ cells exactly have cancer stem cells (CSCs) properties compared to CD44-/ESA- cells. The number of colony and spheroid formed from CSCs was remarkably reduced after carbon ion beam combined with GEM and CDDP compared to that of X-ray or carbon ion beam alone. A more large number and large-sized gamma-H2AX foci were observed in CSCs after 24 h carbon ion beam combined with GEM and CDDP compared to that of X-ray or carbon ion beam alone. High dose of carbon ion beam with 35 Gy alone or with 25 Gy of relative low carbon ion beam combined with GEM and CDDP induced more severe xenograft tumor cell cavitation and fibrosis and significantly regressed the tumors compared to that of X-ray irradiation.
