Colony and spheroid formation assay as well as in vivo tumorigenic assay showed that CD133+/CD90+ or CD44+/ESA+ cells exactly have cancer stem cells (CSCs) properties compared to CD133-/CD90- or CD44-/ESA- cells. The number of colony and spheroid formed from CSCs was remarkably reduced after carbon ion beam combined with sorafenib compared to that of X-ray or carbon ion beam alone. Carbon ion beam combined with sorafenib significantly increased apoptosis-related gene Bax expression, but reduced autophagy-related gene p62 expression compared to that of X-ray or carbon ion beam alone. High dose of carbon ion beam with 35 Gy alone or with 25 Gy of relative low carbon ion beam combined with sorafenib induced more severe xenograft tumor cell cavitation and fibrosis and significantly regressed the tumors compared to that of X-ray irradiation.
