However, the survival rate of the patients is not satisfied because of the distant metastasis by cancer cells that scattered from the primary tumor before radiotherapy. In addition, the effects of carbon-ion beams to metastatic abilities are not clear. The aim of this study are 1) to investigate the effect of a inhibitor of Hsp90, related with the number of metastasis-associated genes, in combination with radiations, and 2) to evaluate the effect of ATP which is released out of the cells after low-dose irradiation to metastatic potentials. The Hsp90 inhibitor, PU-H71 decrease the survival of LM8 cells until 1 μM with the increase of the drug concentration, whereas the effect of the drug was saturated more than 1 μm. PU-H71 showed radiosensitization effects for not only X-rays but also C-ions with high LET (50 keV/μm). Enhancement ratio (E.R.) of PU to X-rays were smaller with increasing drug concentration, however the value to C-ions showed almost constant regardless of the concentration. After the low-dose radiation (0.5 Gy) increased the migration activity of B16/BL6 cells and the degree of the migration after irradiation were enhanced in ATP rich condition, but not ATP depletion condition. In addition, ATP rich condition increased the frequency of γ-H2AX foci, the marker of DNA double strand breaks.}, pages = {150--155}, title = {局所制御と転移抑制を目指した放射線との併用療法の検討}, year = {2013} }
