There are only a few studies in the molecular biological level and gene expression in association with PLD repair, and on limiting its effect after carbon-ion beam irradiation to cancer cells in vivo, no study was reported. In this study we tried to detect the mechanism of DNA damage and repair of the IA tumor cell after carbon-ion beam or X-ray irradiation. At first, colony assay method was performed to compare the time dependent cell survival of the IA cells after each irradiation. Second, to search the genes causing PLD repair after each irradiation, we evaluated the time dependent gene expression by using semi-quantitative RT-PCR with the selected 33 genes reported to be related to DNA repair. From the result of survival assay, PLD repair was found only after irradiation of X-ray in this IA cells, whereas it was inhibited after carbon-ion beam irradiation. Genes showing significantly higher expressions after X-ray irradiation than those after carbon-ion beam irradiation were 5 genes and lower expressions after X-ray irradiation than those after carbon-ion beam irradiation were 7 genes.
