Cell suspension with the concentration of 5x10^7 /ml was irradiated under air-saturated condition. After irradiation cells were subjected to the DNA extraction using isopropanol, separation of DNA strands by heat treatment, digestion into nucleosides with nuclease P1 and alkaline phosphatase. Complete separation into single strands using extensive heating resulted in the formation of a sole peak of 8-OHdG on ECD profile separated by HPLC. Although the preliminary results showed a slight increase in 8-OHdG generation with increasing LET, the reproducibility was rather poor. For an alternative method detection by flow cytometry using FITC-labeled antibody against 8-OHdG was explored. The sensitivity for the detection by the two methods was nearly the same, both of which could detect 8-OHdG after the exposure of 150 Gy of carbon beam with an LET of 20 keV/um.
