Irradiated cells were immuno-stained with gamma-H2AX antibody to visualize the distribution of DNA double strand break (dsb), which specifically induces phosphorylation of H2AX (gamma-H2AX). In an irradiated cell, bright and well-defined fluorescent foci were seen in the nucleus, but in a non-irradiated cell, no or few foci were detected. Cross-sectional views that were constructed with the software using 20 sliced images of a nucleus with 0.55 um interval, showed rod-like shapes of green fluorescence from top to bottom of the nucleus. These must be cross-sectional images of the gamma-H2AX column produced by ions traversing the nucleus. The correlation between the number of gamma-H2AX foci observed and average number of ion traversals through a nucleus showed 0.92 +/- 0.09.
