We used p53 null H1299 cells transfected with wild-type p53(wtp53) or mutated p53(mp53) gene. We found that (i) there was no significant difference in sensitivity to high LET radiations above 30 KeV/um among H1299, H1299/mp53 and H1299/wtp53 cells, although the radiosensitivity of H1299/wtp53 cells against X-rays was higher (1.2-fold) than those of H1299 and H1299/mp53 cells; (ii) there was an synergistic enhancement with heat to radiosensitivity against below 70 KeV/um in H1299/ wtp53 cells alone; (iii) low-LET radiations effectively induced apoptosis in H1299/wtp53 cells as compared with H1299 and H1299/mp53 cells; (iv) high-LET radiations as compared with low-LET radiation at iso-survival dose effectively induced apoptosis in H1299/wtp53 cells; (v) high-LET radiations induced p53-independent apoptosis in H1299 and H1299/mp53 cells; (vi) thermal enhancement of cellular sensitivity against X-ray might be due to induction of p53-dependent apoptosis.
