Carbon ions has been used to treat more than 1000 patients in our institute. One of the tumors responding well to carbon ions is malignant melanomas while squamous cell carcinomas show response comparable to photon radiotherapy. It is possible that the two types of tumors, i.e., malignant melanoma and squamous cell carcinoma, are different in radiosensitivity to carbon ions, provided that the clinical difference between the two tumors is due to any biological reasons. We assumed two possibilities to reason the biological difference between the tumors. First, malignant melanomas in general are more resistant to photons than squamous cell carcinomas while relative biological effectiveness (RBE) of carbon ions are the same between the two tumors. In this case, biological factor(s) that determine radiosensitivities of tumors are independent of radiation qualities. Second, malignant melanomas respond better than squamous cell carcinomas such that RBE of carbon ions are larger for malignant melanomas than squamous cell carcinomas. We will use 10 malignant melanomas and 10 squamous cell carcinomas of human origin, and compare radiosensitivities of each cell line in vitro and in vivo. The ions we use here are both lighter and heavier than carbon ions. In our previous studies of carbon ions, intermediate LET is not only superior to low LET but also more than comparable to high LET in achieving the biological therapeutic gain that is calculated by comparing RBE values between mice skin reaction scoring and tumor growth delay.
