In this study, we examined the radio-sensitivity for carbon ions of defective cell lines in DNA-PKcs (DNA-PK catalytic subunit), ATM or NBS1 and of cells treated with wortmannin, an inhibitor of PI3-kinase, DNA-PK and ATM. We found that DNA-PKcs-, ATM- or NBS1-defective cell lines were more sensitive to both X-rays and carbon ions than wild-type cell lines. The RBEs determined by D10 of carbon ions relative to X-rays were about 2.0 in the defective cell lines. On the other hand, RBEs of wortmannin treated or untreated wild-type cells (LM217 or HeLa cells) were 1.59 or 1.29 (with 10 or 20 uM wortmannin) and 1.95 or 2.17 (untreated) respectively.
