The LET dependences of cell killing on the knockout mutant cells of DNA-dsb repair genes (ku70-/-, rad54-/-, ku70-/-rad54-/-, and the parent DT40), and that of PLDR and SLDR in HSG cells were investigated. LET dependencies of repair from DNA-dsb in DT40 cells showed a big difference revealed by the difference in presence or absence of sister chromatid and repair genes in the cell. A usual LET-RBE relationship was observed in repair sufficient cells, but no peak of RBE was found in repair deficient cells. This suggests that the peak of the RBE caused by misrepair of DNA dsb. LET dependency of SLDR on HSG cells determined by X-ray following exposure of different high-LET as primary irradiation was difficult to find.
