Radiation exposure during the fetal period leads\nto disturbances in development of the brain and can induce\nmicrocephaly or anencephalia. Here we investigated biological\neffects of the X-irradiation on the neural stem cells, prepared from\nmouse embryonic stem (ES) cells by the Neural Stem Sphere (NSS)\nmethod. Cultured with mitogen fibroblast growth factor-2, the\nneural stem cells proliferated exponentially. When the neural stem\ncells were exposed to various doses of X-ray, the proliferation of the\nneural stem cells was dose-dependently inhibited by X-irradiation\nand almost completely stopped by 5 Gy-radiation. Morphology of\nthe multipolar neural stem cells was affected by the radiation, and\nthe surviving cells after radiation were bipolar and larger than the\nneural stem cells. However, immunofluorescent study showed that\nthe surviving bipolar cells were positive for nestin, and gene\nexpression of nestin and musashi-1 was also demonstrated in the\ncells by RT-PCR study.
