A Tetrahymena group I intron ribozyme reporter was split into two halves, and in the presence of the target RNA, the split reporter assembled into an active complex that trans-spliced and produced a reporter enzyme. We show that this split reporter can sense tumor suppressor p53 mutant mRNA in mammalian cells.
