In aggressive ATL, the response to chemotherapy is extremely poor. We hypothesized that this poor response is due to the existence of chemotherapy-resistant cells, such as leukemic stem cells. Previously, we successfully identified an ATL stem cell (ATLSC) candidate as the c-kit+/CD38−/CD71− cells in an ATL mouse model using Tax transgenic mice. Here, with a new ATL mouse model using\nHBZ-transgenic mice, we further discovered that the functional ATLSC candidate,\nwhich commonly expresses c-kit, is drug-resistant and has the ability to initiate tumors and reconstitute lymphomatous cells. We characterized the ATLSCs as c-kit+/CD4−/CD8− cells and found that they have a similar gene expression profile as T cell progenitors. Additionally, we found that AP-1 gene family members, including\nJunb, Jund, and Fosb, were up-regulated in the ATLSC fraction. The results of an in vitro assay showed that ATLSCs cultured with cytokines known to promote stem\ncell expansion, such as stem cell factor (SCF), showed highly proliferative activity and maintained their stem cell fraction. Inhibition of c-kit–SCF signaling with the\nneutralizing antibody ACK2 affected ATLSC self-renewal and proliferation. Experiments in Sl/Sld mice, which have a mutation in the membrane-bound c-kit ligand, found that ATL development was completely blocked in these mice. These results clearly suggest that the c-kit–SCF signal plays a key role in ATLSC self-renewal and in ATL initiation and disease progression.","subitem_description_type":"Abstract"}]},"item_8_publisher_8":{"attribute_name":"出版者","attribute_value_mlt":[{"subitem_publisher":"Impact Journals, LLC"}]},"item_8_relation_14":{"attribute_name":"DOI","attribute_value_mlt":[{"subitem_relation_type_id":{"subitem_relation_type_id_text":"10.18632/oncotarget.10210","subitem_relation_type_select":"DOI"}}]},"item_8_relation_17":{"attribute_name":"関連サイト","attribute_value_mlt":[{"subitem_relation_name":[{"subitem_relation_name_text":"http://www.impactjournals.com/oncotarget/index.php?journal=oncotarget&page=article&op=view&path%5B%5D=10210"}],"subitem_relation_type_id":{"subitem_relation_type_id_text":"http://www.impactjournals.com/oncotarget/index.php?journal=oncotarget&page=article&op=view&path%5B%5D=10210","subitem_relation_type_select":"URI"}}]},"item_8_source_id_9":{"attribute_name":"ISSN","attribute_value_mlt":[{"subitem_source_identifier":"1949-2553","subitem_source_identifier_type":"ISSN"}]},"item_access_right":{"attribute_name":"アクセス権","attribute_value_mlt":[{"subitem_access_right":"metadata only access","subitem_access_right_uri":"http://purl.org/coar/access_right/c_14cb"}]},"item_creator":{"attribute_name":"著者","attribute_type":"creator","attribute_value_mlt":[{"creatorNames":[{"creatorName":"Kuribayashi, Wakako"}],"nameIdentifiers":[{"nameIdentifier":"480497","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Takizawa, Kazuya"}],"nameIdentifiers":[{"nameIdentifier":"480498","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Sugata, Kenji"}],"nameIdentifiers":[{"nameIdentifier":"480499","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Kuramitsu, Madoka"}],"nameIdentifiers":[{"nameIdentifier":"480500","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Momose, Haruka"}],"nameIdentifiers":[{"nameIdentifier":"480501","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Sasaki, Eita"}],"nameIdentifiers":[{"nameIdentifier":"480502","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Hiradate, Yuki"}],"nameIdentifiers":[{"nameIdentifier":"480503","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Furuhata, Keiko"}],"nameIdentifiers":[{"nameIdentifier":"480504","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Asada, Yoshihisa"}],"nameIdentifiers":[{"nameIdentifier":"480505","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Iwama, Atsushi"}],"nameIdentifiers":[{"nameIdentifier":"480506","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Matsuoka, Masao"}],"nameIdentifiers":[{"nameIdentifier":"480507","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Mizukami, Takuo"}],"nameIdentifiers":[{"nameIdentifier":"480508","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Hamaguchi, Isao"}],"nameIdentifiers":[{"nameIdentifier":"480509","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"滝澤 和也","creatorNameLang":"en"}],"nameIdentifiers":[{"nameIdentifier":"480510","nameIdentifierScheme":"WEKO"}]}]},"item_language":{"attribute_name":"言語","attribute_value_mlt":[{"subitem_language":"eng"}]},"item_resource_type":{"attribute_name":"資源タイプ","attribute_value_mlt":[{"resourcetype":"journal article","resourceuri":"http://purl.org/coar/resource_type/c_6501"}]},"item_title":"Impact of the SCF signaling pathway on leukemia stem cell mediated ATL initiation and progression in an HBZ transgenic mouse model","item_titles":{"attribute_name":"タイトル","attribute_value_mlt":[{"subitem_title":"Impact of the SCF signaling pathway on leukemia stem cell mediated ATL initiation and progression in an HBZ transgenic mouse model"}]},"item_type_id":"8","owner":"1","path":["1"],"pubdate":{"attribute_name":"公開日","attribute_value":"2017-04-27"},"publish_date":"2017-04-27","publish_status":"0","recid":"47860","relation_version_is_last":true,"title":["Impact of the SCF signaling pathway on leukemia stem cell mediated ATL initiation and progression in an HBZ transgenic mouse model"],"weko_creator_id":"1","weko_shared_id":-1},"updated":"2023-05-15T23:34:44.972280+00:00"}
