To detect abnormal epigenetic alterations in DNA methylation, convenient and cost-effective\nmethods are required for such research, in which multiple samples are processed simultaneously. We here present\nmethylated site display (MSD), a unique technique for the preparation of DNA libraries. By combining it with amplified\nfragment length polymorphism (AFLP) analysis, we developed a new method, MSD-AFLP.\nResults: Methylated site display libraries consist of only DNAs derived from DNA fragments that are CpG methylated\nat the 5′ end in the original genomic DNA sample. To test the effectiveness of this method, CpG methylation levels in\nliver, kidney, and hippocampal tissues of mice were compared to examine if MSD-AFLP can detect subtle differences\nin the levels of tissue-specific differentially methylated CpGs. As a result, many CpG sites suspected to be tissue-specific\ndifferentially methylated were detected. Nucleotide sequences adjacent to these methyl-CpG sites were identified\nand we determined the methylation level by methylation-sensitive restriction endonuclease (MSRE)-PCR analysis\nto confirm the accuracy of AFLP analysis. The differences of the methylation level among tissues were almost identical\namong these methods. By MSD-AFLP analysis, we detected many CpGs showing less than 5% statistically significant\ntissue-specific difference and less than 10% degree of variability. Additionally, MSD-AFLP analysis could be used to\nidentify CpG methylation sites in other organisms including humans.\nConclusion: MSD-AFLP analysis can potentially be used to measure slight changes in CpG methylation level. Regarding\nthe remarkable precision, sensitivity, and throughput of MSD-AFLP analysis studies, this method will be advantageous\nin a variety of epigenetics-based research.\nKeywords: DNA methylation profiling, AFLP, Epigenetics","subitem_description_type":"Abstract"}]},"item_8_publisher_8":{"attribute_name":"出版者","attribute_value_mlt":[{"subitem_publisher":"BioMed Central "}]},"item_8_relation_14":{"attribute_name":"DOI","attribute_value_mlt":[{"subitem_relation_type_id":{"subitem_relation_type_id_text":"10.1186/s12867-017-0083-2","subitem_relation_type_select":"DOI"}}]},"item_8_relation_17":{"attribute_name":"関連サイト","attribute_value_mlt":[{"subitem_relation_name":[{"subitem_relation_name_text":"https://www.ncbi.nlm.nih.gov/pmc/articles/PMC5345256/pdf/12867_2017_Article_83.pdf"}],"subitem_relation_type_id":{"subitem_relation_type_id_text":"https://www.ncbi.nlm.nih.gov/pmc/articles/PMC5345256/pdf/12867_2017_Article_83.pdf","subitem_relation_type_select":"URI"}}]},"item_8_source_id_9":{"attribute_name":"ISSN","attribute_value_mlt":[{"subitem_source_identifier":"1471-2199","subitem_source_identifier_type":"ISSN"}]},"item_access_right":{"attribute_name":"アクセス権","attribute_value_mlt":[{"subitem_access_right":"metadata only access","subitem_access_right_uri":"http://purl.org/coar/access_right/c_14cb"}]},"item_creator":{"attribute_name":"著者","attribute_type":"creator","attribute_value_mlt":[{"creatorNames":[{"creatorName":"Aiba, Toshiki"}],"nameIdentifiers":[{"nameIdentifier":"478912","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Saito, Toshiyuki"}],"nameIdentifiers":[{"nameIdentifier":"478913","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Hayashi, Akiko"}],"nameIdentifiers":[{"nameIdentifier":"478914","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Sato, Shinji"}],"nameIdentifiers":[{"nameIdentifier":"478915","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Yunokawa, Harunobu"}],"nameIdentifiers":[{"nameIdentifier":"478916","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Maruyama, Toru"}],"nameIdentifiers":[{"nameIdentifier":"478917","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Fujibuchi, Wataru"}],"nameIdentifiers":[{"nameIdentifier":"478918","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Kurita, Hisaka"}],"nameIdentifiers":[{"nameIdentifier":"478919","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Tohyama, Chiharu"}],"nameIdentifiers":[{"nameIdentifier":"478920","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Ohsako, Seiichiroh"}],"nameIdentifiers":[{"nameIdentifier":"478921","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"相場 俊樹","creatorNameLang":"en"}],"nameIdentifiers":[{"nameIdentifier":"478922","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"齋藤 俊行","creatorNameLang":"en"}],"nameIdentifiers":[{"nameIdentifier":"478923","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"林 昭子","creatorNameLang":"en"}],"nameIdentifiers":[{"nameIdentifier":"478924","nameIdentifierScheme":"WEKO"}]}]},"item_language":{"attribute_name":"言語","attribute_value_mlt":[{"subitem_language":"eng"}]},"item_resource_type":{"attribute_name":"資源タイプ","attribute_value_mlt":[{"resourcetype":"journal article","resourceuri":"http://purl.org/coar/resource_type/c_6501"}]},"item_title":"Methylated site display (MSD)-AFLP, a sensitive and affordable method for analysis of CpG methylation profiles","item_titles":{"attribute_name":"タイトル","attribute_value_mlt":[{"subitem_title":"Methylated site display (MSD)-AFLP, a sensitive and affordable method for analysis of CpG methylation profiles"}]},"item_type_id":"8","owner":"1","path":["1"],"pubdate":{"attribute_name":"公開日","attribute_value":"2017-04-20"},"publish_date":"2017-04-20","publish_status":"0","recid":"47729","relation_version_is_last":true,"title":["Methylated site display (MSD)-AFLP, a sensitive and affordable method for analysis of CpG methylation profiles"],"weko_creator_id":"1","weko_shared_id":-1},"updated":"2023-05-15T23:36:11.105044+00:00"}
