The structurally\ndisordered N-terminal tail of DDB2 contains\nseven lysines identified as major sites for ubiquitination\nthat target the protein for proteasomal degradation;\nhowever, the precise biological functions of\nthese modifications remained unknown. By exogenous\nexpression of mutant DDB2 proteins in normal\nhuman fibroblasts, here we show that the N-terminal\ntail of DDB2 is involved in regulation of cellular responses\nto UV. By striking contrast with behaviors\nof exogenous DDB2, the endogenous DDB2 protein\nwas stabilized even after UV irradiation as a function\nof the XPC expression level. Furthermore, XPC competitively\nsuppressed ubiquitination of DDB2in vitro,\nand this effect was significantly promoted by centrin-\n2, which augments the DNA damage-recognition activity\nof XPC.
