Both pre- and\npostsynaptic dopaminergic functions can be estimated by positron\nemission tomography (PET) with different radiotracers. However, an\nintegrated database of both pre- and postsynaptic dopaminergic\nneurotransmission components including receptors, transporter, and\nendogenous neurotransmitter synthesis has not yet been reported. In the\npresent study, we constructed a normal database for the pre- and\npostsynaptic dopaminergic functions in the living human brain using\nPET. To measure striatal and extrastriatal dopamine D1 and D2 receptor\nbindings, dopamine transporter binding, and endogenous dopamine\nsynthesis rate, PET scans were performed on healthy men after intravenous\ninjection of [11C]SCH23390, [11C]raclopride, [11C]FLB457,\n[11C]PE2I, or L-[β-11C]DOPA. All PET images were anatomically\nstandardized using SPM2, and a database was built for each radiotracer.\nGray matter images were segmented and extracted from all anatomically\nstandardized magnetic resonance images using SPM2, and they\nwere used for partial volume correction. These databases allow the\ncomparison of regional distributions of striatal and extrastriatal\ndopamine D1 and D2 receptors, dopamine transporter, and endogenous\ndopamine synthesis capability. These distributions were in good\nagreement with those from human postmortem studies. This database\ncan be used in various researches to understand the physiology of\ndopaminergic functions in the living human brain. This database could\nalso be used to investigate regional abnormalities of dopaminergic\nneurotransmission in neuropsychiatric disorders.\n© 2007 Elsevier Inc.
